Albert staining
Albert's staining is usually used for metachromatic granules present in certain bacilli such as;
Corynebacterium
Diphtheria
Lactobacilli
Procedure of Albert's Staining
1. Prepare smear and fix it.
2. Cover with Albert's stain for 5 minutes.
3. Wash under water
4. Cover smear with Albert's iodine solution before 2 minutes.
5. Wash under water and dry it.
6. Examine in oil immersion under microscope.
Observation and Report
Metachromatic granules stain bluish black and bacterial protoplasm stain green.
